DIBES (Drebrin-imaging-based Evaluation of mature Synapses)
In this section, we introduce the easiest method by which the accumulation of drebrin at dendritic spines is reproducibly and quantitatively.
As a marker of mature synapses, the appearance of slow tread-milling, stable actin filaments with 40 nm pitch is most appropriate. Because the stable actin inculde drebrin, the accumulation of drebrin at dendritic spines is a good surrogate marker for the appearance of stable actin filaments. Using drebrin immunopositive clusters along dendrite r, the nuber of mature synapses is easily measured.
